Epilepsy Drugs Linked to Birth Defects. A new study of antiepileptic drugs (AEDs) has found that pregnant women taking such medications were more likely to give birth to children with certain birth defects. The AEDs birth defect study, which was recently presented at the 62nd Annual Meeting of the American Epilepsy Society, found that valproate was strongly associated with an increased risk of birth defects, while other AEDs such as lamotrigine and carbamazepine also showed an increased risk.

To compare different AEDs with incidence of  birth defects, scientists conducting this research retrospectively analyzed data from epilepsy and pregnancy registries, including the North American Antiepileptic Drug Pregnancy Registry, the UK Epilepsy and Pregnancy Registry, the Lamotrigine Pregnancy Registry, the Finnish Drug Prescription database, the Swedish Medical Birth registry, and the Australian AED and Pregnancy Registry.

Overall, the study found that the prevalence of birth defects among babies born to women taking AEDs was about 6%, whereas the prevalence of birth defects in the general population is about 0.2%, similar to the rate among women with epilepsy who are not taking AEDs. In addition, the analysis found that  treatment with more than one AEDs had  a higher malformation rate than monotherapy with AEDs (6.8% vs 4%).

highest malformation rate was associated with valproate

In the North American registry, the highest malformation rate was associated with valproate (10.7%), followed by phenobarbital (6.5%) and lamotrigine (2.3%). The rate for lamotrigine was comparable to that of carbamazepine. In addition, valproate had a malformation rate that was about seven times higher than the external control population. Phenobarbital had a malformation rate five to six times higher than the control population. Lamotrigine and carbamazepine rates were not significantly higher than control population rates.

Sixteen major malformations occurred in the offspring of 149 women who used valproate during pregnancy, and these malformations included neural tube defects, craniofacial defects, cardiovascular malformations and malformations involving other body systems.

In the UK Epilepsy and Pregnancy Register, the malformation rate with valproate was 6.3%, whereas lamotrigine and carbamazepine malformation rates were significantly lower. The Lamotrigine Pregnancy Registry, the Finnish Drug Prescription database, the Swedish Medical Birth registry, and the UK, North American, and Australian AED and pregnancy registries all showed a pattern of high malformation rates with valproate.

U.S.FDA reminded health care providers about the increased risk of neural tube defects

Last week, the U.S. Food & Drug Administration (FDA)  reminded health care providers about the increased risk of neural tube defects and other major birth defects, such as craniofacial defects and cardiovascular malformations, in babies exposed to valproate sodium products during pregnancy.  The agency advised that women only use valproate if it is essential to manage their medical condition. Women not actively planning a pregnancy should use effective contraception if they are taking valproate, as birth defect risks are particularly high during the first trimester, before many women know they are pregnant.

The FDA said it will be working with the manufacturers of valproate products to address labeling changes.

In the U.S., valproate sodium is marketed as Depacon. Dilvalproex sodium is marketed as Depakote, Depakote CP, Depakote ER. Valproic acid is marketed as Depakene and as Stavzor.

Parker Waichman LLP- Paralegal, Jessie Salcedo, she is professional and has been very helpful. I am a new mom and a newlywed so my life is very chaotic at times. Sometimes it's hard todo anything else but my basic day to day stuff. They have made this process very easy for me. All I've really had todo is provide some information and they have handled everything else. I greatly appreciate their services and would refer them to anyone. Also, they remembered my birthday this past September. They sent me a card. Most of my friends don't remember my birthday. To me, that was going out of their way just to brighten my day and I really appreciated it.

When I signed up with the Victim's Compensation Fund ("VCF"), Parker Waichman ("PW") guided me through the entire process. Without their guidance I wouldn't have gotten through it. They do outstanding work and they care deeply for their clients. Tina Morace is pleasant, patient, and very professional. She held my hand through the process of getting all the documents together. Tina is a valuable employee at PW, I hope PW appreciates her as much as I do.
